Overview

DL-Dithiothreitol (DTT) is a well-known reducing agent used in biochemistry and molecular biology approaches. DTT penetrates the cellular membrane to safeguard protein sulfhydryl groups in reduced state, consequently restoring enzyme activity that may have been compromised due to oxidation of these functional groups.
Dithiothreitol can act as a less toxic substitute for 2-mercaptoethanol in most applications; typically, a seven-fold lower concentration of DTT (100 mM) is needed than is used for 2-mercaptoethanol (700 mM).
The optimal pH range for DTT is 7.1-8.0. It is less potent reducing agent with decreasing pH levels (< pH 6.5).
