L-Glutamine 100x is a concentrated sterile solution of the essential amino acid L-glutamine, used as a supplement in cell culture medium. It plays a key role in cellular metabolism, serving as a major source of nitrogen and carbon, which are essential for synthesizing proteins, nucleotides, and other molecules required for cell proliferation. In addition, L-glutamine is critical for maintaining cell viability, especially in fast-growing cells like hybridomas, Chinese Hamster Ovary (CHO) cells, and various cancer cell lines.
The concentration of L-glutamine needed varies depending on the cell type and medium formulation. Classical cell culture media like DMEM, GMEM, and IMDM typically contain L-Glutamine concentrations ranging from 2 mM to 4 mM, which are adequate for most cell lines. However, certain cells, such as hybridoma cells, may require higher concentrations for optimal growth.
In specialized media, such as serum-free or protein-free formulations, L-glutamine is often supplemented at levels around 2.5 mM to 6 mM.
However, L-Glutamine, unlike other amino acids, is unstable in solution. It tends to degrade over time, particularly under conditions of elevated temperature and neutral to basic pH. This degradation results in the formation of toxic by-products, such as ammonia, which can negatively affect cell growth and viability. Therefore, fresh L-glutamine is often added to cell culture media, or more stable alternatives like L-alanyl-L-glutamine (a dipeptide form) are used to overcome this instability.
Since L-glutamine degrades in solution, researchers need to carefully manage its concentration and storage.
