Proteinase K is a serine protease with broad cleavage specificity. The enzyme, isolated from the fungus Tritirachium album is produced as recombinant protein. It is used in DNA and RNA extraction protocols to eliminate proteins.
Proteinase K is a robust serine protease with broad substrate specificity, capable of efficiently degrading a wide range of proteins, including those in their native conformation. It remains active in the presence of common detergents, such as 1% Triton X-100 and 0.5% (w/v) SDS.
In molecular biology, Proteinase K is widely used to remove contaminating proteins, including nucleases, during the purification of DNA and RNA from microorganisms, cultured cells, and plant tissues. It is also effective in eliminating endotoxins associated with cationic proteins such as lysozyme and ribonuclease A, thereby improving the quality of nucleic acid preparations.
